Can a timing chain be tightened?

You need a gasket between the block and timing chain cover, and I used liquid gasket from AutoZone for the water pump. Also, you should use teflon tape or thread sealer on the water pump bolts that go into the block because they will leak otherwise.

I've always been advised against using rtv sealant AND a gasket. But the manual says to put some sealer on the gasket. So really put a thin layer of rtv sealant on the gaskets where it states that (ie water pump and timing chain cover)? Or would it be better to only use the rtv sealant as was also mentioned?

I'd just go with the rtv. Otherwise you have a completely watertight seal on one side, and a somewhat watertight seal on the other. The gasket will fail before the rtv. Also, make sure that all your hoses are as tight as you can get them. Otherwise, they will leak.

So rtv on the water pump, the gasket that came with the cover, with a thin layer of rtv on both sides (thread sealer on the bolts), and the rubber gasket on the oil pan?

I also have a rubber gasket for the tranny pan, and a paper type gasket for the rear end.
