I think I have a slight misfire...
Can I check for a misfire by pulling off the coil packs one by one and then replacing them while it's running?

Not a good idea. Why?

Without spark a full load of raw fuel will be dumped into the exhaust. This may damage the cats.

If the COP secondary arcs it may damage the insulation. If there happens to be a body part in the way, have a buddy video it. It might make a good "Max-X" segment.

Better idea is to look at the DTC code. This will often tell you which cylinder is misfiring.

If no particular cylinder is misfiring, suggest looking at the EGR system as a possble cause.

If there really is a need to disable a cylinder, disconnect the fuel injector instead.
